Help with .mo translation

I'd be grateful for some advice on the following:
I want to change the site text (not change language) to reflect the nature of the site so I've downloaded the UK .po file and done the following:
Changed required phrases and words using poedit
exported a .mo file
made a folder called languages in
wp-content
re-named file to en_UK.mo and uploaded to languages folder
in
wp-config typed this line - define ('WPLANG', 'en_UK');
My translated .mo file is not showing e.g. words and phrases I changed are not apparent on the site - I wondered have I done everything?
I realise it's a bit 'off-message' but any advice thankfully received
